Genipin, an aglycon of geniposide, has been reported to exhibit diverse pharmacological functions such as antitumor and anti-inflammatory effects. This study aimed to elucidate the anti-inflammatory mechanism of genipin, focusing particularly on the role of heme oxygenase-1 (HO-1), a potent anti-inflammatory enzyme. In RAW264.7 cells, genipin increased HO-1 expression and its enzyme activity via a NF-E2-related factor 2 (Nrf2)–antioxidant response element (ARE) pathway. These effects were significantly inhibited by exposure to the phosphatidylinositol 3-kinase (PI 3-kinase) inhibitor, LY294002, or by expression of a dominant negative mutant of PI 3-kinase. Additional experiments showed that the activation of c-Jun NH2-terminal kinase 1/2 (JNK1/2) is required for genipin-induced phosphorylation and nuclear translocation of Nrf2 and antioxidant response element (ARE)-driven induction of HO-1, and acts as a downstream effector of PI 3-kinase. Furthermore, functional significance of HO-1 induction was revealed by genipin-mediated inhibition of lipopolysaccharide-stimulated inducible nitric oxide synthase expression or cyclooxygenase-2 promoter activity, the response was reversed by the blocking of HO-1 protein synthesis or HO-1 enzyme activity. Therefore, identification of PI 3-kinase-JNK1/2-Nrf2-linked signaling cascade in genipin-mediated HO-1 expression defines the signaling event that could participate in genipin-mediated anti-inflammatory response.  相似文献

Perchlorate (ClO4-) contamination of groundwater has recently become a major concern across the nation. Electrokinetic (EK) extraction with the simultaneous EK injection of organic material to promote degradation could allow for the efficient removal of perchlorate while simultaneously promoting degradation of perchlorate. Column experiments were conducted to evaluate the technology. Lactate and glycine served as organic substrates to promote degradation after injection into the columns as well as maintaining the pH near neutral. Removal of perchlorate from contaminated materials kaolin, sand, and a natural soil historically contaminated by perchlorate was controlled by the ionic flux of perchlorate and not by transport from the osmotic flux which was only significant for kaolin experiments. Perchlorate was removed from contaminated sand and clay below our detection limits (5 ppb). Both lactic acid and glycine were successfully injected into clay and a sand matrix. Results from a contaminated site soil indicate that the Chemical Oxygen Demand was increased after electrokinetic injection of glycine and lactate. Experiments using soil from a contaminated site confirmed that EK can be used to both remove perchlorate and stimulate bioremediation by the injection of lactate or glycine. The use of EK technology to both remove and provide for continued source removal by bioremediation offers a potential new tool to treat low permeability systems.  相似文献

A maize transposable family, Ac/Ds, has been successfully utilized as a mutagenizing agent not only in monocot but also in dicot. In order to develop insertional mutagenesis system in pepper, the mobility of Ac/Ds has been examined. In this study, the excision of the elements was monitored via transient assay system with protoplasts. Two different systems were developed and compared; one- and two-elements systems. In a one-element system, Ac alone was introduced into cells. As a two-element system, Ac and Ds were cloned into a single vector and were expressed in protoplasts. Our data showed that both Ac and Ds elements were highly mobile in pepper cells. This is the first report suggesting that Ac/Ds mediated gene tagging system could be successfully operated in pepper.  相似文献

Background

Acute respiratory distress syndrome (ARDS) is potentially underrecognized by clinicians. Early recognition and subsequent optimal treatment of patients with ARDS may be facilitated by usage of biomarkers. Surfactant protein D (SP-D), a marker of alveolar epithelial injury, has been proposed as a potentially useful biomarker for diagnosis of ARDS in a few studies. We tried to validate the performance of plasma SP-D levels for diagnosis of ARDS.

Methods

We conducted a retrospective analysis using data from three (two in USA and one in Korea) prospective biobank cohorts involving 407 critically ill patients admitted to medical intensive care unit (ICU). A propensity score matched analysis (patients with versus without ARDS, matched 1:1) was carried out using significant variables from multiple logistic regression. The diagnostic accuracy of plasma SP-D as a diagnostic marker of ARDS was assessed by receiver operating characteristic curve analysis.

Results

Out of the 407 subjects included in this study, 39 (10%) patients fulfilled ARDS criteria. Patients with ARDS had higher SP-D levels in plasma (p?<?0.01) and higher hospital-mortality (p?<?0.001) than those without ARDS. Thirty eight subjects with ARDS (cases) were successfully matched for propensity for ARDS with 38 subjects without ARDS (controls). Plasma levels of SP-D were higher in cases with ARDS compared to their matched controls without ARDS [median 20.8 ng/mL (interquartile range, 12.7–38.4) versus 7.9 (4.1–17.0); p?=?0.001]. The area under the receiver operating characteristic curve for SP-D for the diagnosis of ARDS was 0.71 (95% confidence intervals, 0.60–0.83). A cut-off point of 12.7 ng/mL for SP-D yielded sensitivity of 74% and specificity of 63%.

Conclusions

High levels of SP-D within 48 h after ICU admission might serve as a diagnostic marker for ARDS in patients hospitalized in medical ICU. Further prospective trials are required to validate the diagnostic role of SP-D in ARDS, and if its usefulness is greater in direct than in indirect ARDS, as well as across different strata of severity of ARDS.

spinster (spin) is a late endosome/lysosome membrane protein with the amino acid sequence of a lysosomal sugar carrier and expressed in the glial cells. Spin is required for autophagy and lysosome reformation by releasing lysosomal degradation products of autolysosome into the cytosol in Drosophila larvae and adults. However, such kind of function has not been investigated in embryos yet. In this study, for the first time, we examined the effects of spin mutation on the endocytic pathway and autophagy during embryogenesis. Loss-of-function spin mutation led to the abnormal process of early endosome/recycling endosome and the accumulation of enlarged autophagosome/autolysosome. These abnormal endocytic pathway and autophagy subsequently caused the malformation of head at embryonic stages. These results show that Spin is involved in the endocytic pathway and autophagy during embryogenesis as well as larval and adult stages.  相似文献

Endemic species typically have a narrow niche breadth, and are likely more vulnerable to extinction than more widespread taxa. Squalidus multimaculatus is a small cyprinid endemic to the Korean Peninsula, and its reported geographical range was restricted to several small rivers located along the southeast coast. Several populations of S. multimaculatus have supposedly been subject to a variety of recent anthropogenic actions. Here, we analyzed the pattern of genetic diversity within and among populations of S. multimaculatus using nine microsatellite loci to quantify the relative contributions of human-mediated processes to the contemporary distribution and genetic structure. Overall, low levels of genetic diversity were exhibited in the populations of S. multimaculatus. Genetic differentiations among populations were not completely represented by their geographical proximity, likely resulting from the low intrapopulation genetic variability and anthropogenic transplants. The most conspicuous outcome of the anthropogenic activities was the introgression of alleles from a closely related species, S. gracilis majimae. Our study showed that anthropogenic transplanting, even with only a small number of individuals, can challenge our conservation goal to maintain the species integrity that has long been shaped in evolutionary processes.  相似文献

In this study, we demonstrate the antibacterial activity of silver nanoparticles (AgNPs), depending on their size and shape, on green fluorescent protein (GFP)-expressing E. coli, which provides a facile, rapid, and noninvasive monitoring system. By measuring optical density and fluorescence intensity in the recombinant E. coli, we found that smaller sized plate-shaped AgNPs presented higher antibacterial activity than larger sized, cubic and spherical AgNPs. In the case of 10 nm spherical AgNPs, the optical density was detectable at 15 ng/mL after 12 h incubation, but the fluorescence intensity was not. On the other hand, smaller-sized AgNPs showed higher toxicity than plate-shaped AgNPs based on the measurement of the optical density and fluorescence intensity. The combined analysis of optical density and fluorescence intensity may be helpful for understanding the effect of various materials, including nano- and organic materials, on recombinant bacteria.  相似文献
